I'm from Louisville, MS (30 Miles SE of Starkville) and I tend to use the simple route (I-65 to Bham, Whatever the Interstate is to Tuscolusa, then Highway 82 to Columbus, MS then to Starkville). This is pretty much the longest possilble way to go, but is mostly interstate and a fairly enjoyable ride. If you are only going there once you may want to take this route (it is, above all, an easy ride). Travel time is about 4.5 hours or so (at legal speeds, and a lunch stop).
There are shorter routes, of course. The shortest time I've heard of is about 2.5 hours. I don't know the exact route, but that was driving (in the driver's own words) "like a bat out of hell."
If I may ask, why are you going there? Visiting Mississippi State University (about the only reason I know of for going there! :-) )? Nice place. Small, but I liked it.
